Sermorelin (GHRH(1-29)NH2) is a synthetic 29-amino acid amidated peptide representing the biologically active N-terminal fragment of endogenous growth hormone-releasing hormone (GHRH). It is the shortest fragment of GHRH retaining full receptor binding and activation capacity at the growth hormone-releasing hormone receptor (GHRHR), a class B G protein-coupled receptor expressed primarily on somatotroph cells of the anterior pituitary. Sermorelin is used in research contexts involving hypothalamic-pituitary somatotropic axis signaling, growth hormone secretagogue pharmacology, and pituitary cell biology.

Sermorelin activates GHRHR via Gαs-coupled cAMP/PKA signaling, leading to downstream activation of growth hormone (GH) gene transcription and secretion from pituitary somatotroph cells in primary culture and cell line models. Research involving sermorelin has examined GHRHR signal transduction kinetics, pituitary somatotroph cell proliferation, and the modulation of GH pulsatility patterns in hypothalamic-pituitary organotypic cultures. It is also studied as a reference agonist for GHRHR receptor binding affinity assays and receptor occupancy studies, and has been used to probe the downstream IGF-1 axis in hepatocyte cell culture systems. Sermorelin’s shorter sequence relative to full-length GHRH(1-44) makes it a tractable research tool for structure-activity relationship studies of GHRHR N-terminal binding domain interactions.
