TB-500 acts through G-actin sequestration via the LKKTET motif, inhibiting actin polymerization, a mechanism studied in the context of cell migration, lamellipodia formation and cytoskeletal remodeling. Published research has examined it in the context of integrin-linked kinase (ILK) pathway activation and its interaction with myocardin-related transcription factor A (MRTF-A), which links actin dynamics to gene expression. The acetylated N-terminus is critical to its actin-binding properties.
